During mitosis, the human’s nuclear DNA is compacted in the famous chromosome X-shape. This had been observed using microscopes for more than a century. However, the mechanics of this fundamental process are still not well understood. In this study, the authors present an experimental workflow that allows them to explore the mechanical organization of human chromosomes, revealing insights into these dynamics (including nonlinear stiffness behavior in high force ranges, and the role of agents like topoisomerase IIα (TOP2A)).
